Molecular genetic analysis of the genes for salinomycin biosynthesis in Streptomyces albus

Abstract
Streptomyces albus wild type produced salinomycin and the strain 14¡­33 chromosomal DNA was used for the construction of genomic library using pKC 505 cosmid vector. The genomic library was screened using act 1 (gene for actinorhodin synthase-condensing enzyme) probe and p22¡­81 was isolated. TLC, HPLC, and MS analyses revealed that S. lividans transformed with the p22¡­81 produced salinomycin. The p22¡­81 contained about 24 kb insert and a region of 4.4 kb fragment of the insert hybridized with actI and ACP (acyl-carrier protein) probes was sequenced. Five ORFs found in the same direction were homologus to tcmI, tcmK, tcmL, tcmM and actIII, respectively (tcm; tetracenomycin, act; actinorhodin), tcmI is a cyclase in tetracenomycin synthase gene clusters and tcmK & tcmL are gene for heterodimeric beta-acyl synthase. tcmM codes ACP and actIII is a gene for ketoreductase.
